The Manifestation of C-erbB-2 and p16 Gene in Colon Cancer and Their Clinical Significance

Abstract


Objective This paper aims to reveal the manifestation of C-erbB-2 gene and the anti-cancer gene p16 in Colon cancer and their relation to the pathological mechanism. This research will also make a discussion for the correlation between the two genes in colon cancer pathology, thereby offer some basis for staging and prognosis of colon cancer.Methods This research applied the ELiVisionTM plus methods to analyze C-erbB-2 and p16 genes of fifty patients with colon cancer. We conducted immunehistochemestry test of C-erbB-2 and p16 on the primary tumor and pathologically testified normal colonic mucosa (beyond the 5cm-perimeter of tumor), and analyzed their manifestation in colon tumors and tumor perimeters, and further checked their positive manifestation rates against factors such as age, gender, size of tumor, lymph node metastasis, tumor infiltration depth, differentiation degree, distant metastasis, Dukes stages.Results The cancer gene C-erbB-2 and its anit-gene p16 both occurs in colon tumors and their adjacent tissue. In tumor tissue, the manifestation rates for C-erbB-2 and p16 are 68% and 42% respectively; in adjacent tissue, the rates are 36% and 74% respectively. The difference is statistically significant(P<0.01). C-erbB-2 occurs more in low or medium differentiational glandular cancers, cancers with lymph node metathesis, tumors with infiltration to the serosa, and DukesC+D(P<0.05) , p16 occurs more in high differentiational glandular cancers, cancers without lymph node metathesis, tumors without infiltration to the serosa, and DukesA+B(P>0.05) , Both have nothing to do with gender, age and tumor size of patients(P>0.05). The two genes are negatively correlated in the development of Colon Cancer.Conclusion A higher occurrence of C-erbB-2 or a lower occurrence of p16 will contribute to the occurrence, development, infiltration and transference of colon cancer. They both are malignant biological characteristics of colon cancer; the study of them facilitates our analysis and prediction of lymph node metastasis, seriousness, and clinic stages of colon cancer. The analysis of the two negatively correlated genes will contributes to the assessment of the illness, and prognosis of colon cancer, thereby increase clinic efficiency.
