Abstract


Human respiratory syncytial virus (human respiratory syncytial virus, HRSV) is a worldwide cause one of the important viral pathogens of lower respiratory tract infections in all age groups, particularly seen in infants and young children, the elderly and immunosuppressed individuals, but as yet no clinical safety effective vaccine. The purpose of the research projects is to filter out one or several the HRSV ideal purification methods, and the purified product immunogenicity evaluation, accumulate some basic information for the HRSV cracking purified vaccine. The study includes: of HRSV in human diploid KMB17 cells multiply, the virus harvest fluid ultrafiltration of HRSV different methods of purification and purification conditions to explore, and immunogenicity of the purified product evaluation, and inactivated , adjuvants and immunization programs and other factors affecting the immunogenicity exploration. The use of cell factories and 10 Roche bottles the two container cultured HRSV both Earn higher infectious titer of the virus. HRSV cell factories culture needs to be explored further. 100kD membrane-liquid package ultrafiltration virus harvest was concentrated 30 times and 103 times, respectively, were purified using sucrose density gradient centrifugation and Sepharose 4FF gel filtration chromatography, and the product was purified infectious titer detected, SDS reduction electrophoresis analysis-PAGE and Western blot identification. By the reduction of the electrophoresis, the purified product prepared by the two kinds of purification methods between 60 to 70kD presented a strip Western blot confirmed the specific HRSV F protein. The results showed that the the the Sepharose 4FF Gel filtration chromatography than sucrose density gradient centrifugation method is more suitable for the purification of HRSV, better and higher efficiency. And purification of the column volume, sample volume, elution flow rate conditions would have an impact. Sucrose density gradient centrifugation and Sepharose 4FF 2 ways gel filtration chromatography purified the HRSV obtained samples immune ICR mice, antibodies, seroconversion rates and geometric mean antibody titer (GMT) similar immunization dose (former virus protein content is 2.5 times the latter), but both the immunogenicity was no significant difference, indicating a higher immunogenicity of the Sepharose 4FF Gel filtration chromatography of the purified product. The immunogenicity of the purified sample Exploration on Factors Affecting formaldehyde inactivated group seroconversion rate and antibody GMT than beta-C inactivated lactone group is high; the aluminum adjuvant experimental vaccine immunogen little effect; Od, 7d, 14d, and 0d, 28d two immunization program could be induced relatively high antibody levels (1:9.51 to 1:11.31), but the results suggest that multiple dose helps to improve seroconversion rate and neutralizing antibody titer.
