Objective: To testify the existence of Mena protein in hepatic carcinoma and to explore the expression of Mena protein in hepatic carcinoma and its potential clinical significance with indirect immunohistochemistry.Methods: 54 cases of hepatic carcinoma were selected from the First Assistant Hospital of Dalian Medical University and Dalian central hospital. The positive expression of Mena protein in hepatic carcinoma was testifid first with indirect immunohistochemistry, and then the whole cases were divided into several groups through age, sex, the average diameter of carcinoma, the focus number of carcinoma, the formation of Portal vein tumor thrombus (PVTT), the level of AFP in blood, the infection of HBV, and the TNM clinical stag of hepatic carcinoma. Using statistics to analysis the different expression of Mena protein in these groups, the statistical significance of different groups can be found.Results: The color of hepatic carcinoma stained with immunohistochemistry is yellow or brown. By immunohistochemical staining score of commonly used standard,none positive cell sample gets 0,and positive cell less than 33% gets 1, and so on, 33%-66% gets 2, more than 66% gets 3. By immunohistochemical staining score of color comparisons. The color closed to normal cells gets 0,and so on, light yellow gets 1, brown madder gets 2,and brown gets 3. Divided by the sum of the two scores of 2, and 0 is negative, 0-2 is weak positive, 2-3 is positive, and 3 is strong positive. 1,6 of the 54 cases were Mena negative and the rest were positive, 9 is weak positive and 20 is positive and 19 is strong positive. Positve rate is 89%. 2,The expression of Mena protein in those cases with diameter above and beyond 5 cm , with portal vein tumor thrombus infiltration and with late clinical stage was higher than those without.(P<0.05) There were on differences of expression of Mena protein between sex ,age, numbers of tumor focuses, AFP, HBsAg.(P>0.05)Conclusions: Mena positive is in most cases. (89%) Mena is localized in the cytoplasm of positive cells. In advanced hepatic carcinoma,some of positive cell appears Mena staining juxtamembrane reinforcement The high expressions of Mena protein may have relationship with the development of hepatic carcinoma.
