Expression and Clinical Significance of HMGA1 and HMGA2 in Rhabdomyosarcoma

Abstract


Objective: To investigate the high mobility group protein A1 (HMGA1) and high mobility protein A2 (HMGA2) expression in rhabdomyosarcoma (rhabdomyosarcoma, RMS) characteristics , and its relationship with clinicopathological parameters , cell proliferation . Methods: Immunohistochemistry (IHC) were detected in 39 cases of rhabdomyosarcoma and 10 normal striated muscle tissue HMGA1 protein levels of HMGA2 and Ki-67 ; situ hybridization (ISH) were detected in 39 cases of rhabdomyosarcoma HMGA1 and HMGA2 mRNA expression . Results : HMGA1 and HMGA2 protein expression in the RMS positive rate were 76.9% ( 30 / 39 ) and 64.1% ( 25 / 39 ) , the expression levels were higher than normal striated muscle tissue ( P lt; 0.05 ) : mRNA expression positive rate were 69.2% ( 27 / 39 ) and 66.7% ( 26 / 39 ) , the expression levels were higher than normal striated muscle tissue ( P lt; 0.05 ) . Has a better consistency between HMGA1, HMGA2 expression at the protein level , their respective mRNA levels of expression . Respectively differential expression in embryonal (ERMS) and alveolar (ARMS) was not statistically significant ( P gt; 0.05 ) expression differences between various degrees of differentiation with statistical significance ( P lt ; 0.05 expression ); recurrent or metastatic cases and cases of recurrence or metastasis difference was statistically significant ( P lt; 0.05 ) ; between Ki-67 -positive high proliferation group with Ki-67 -negative low proliferation group expression of the difference was not statistically significant (P gt; O.05). Conclusion : HMGA1 and HMGA2 overexpression may be closely related to the occurrence and development of the RMS , is expected to become one of the indicators to judge the prognosis of the tumor , and provide the basis for targeted therapy .
