Objective: To detect the cGMP-dependent protein kinase Ⅱ (cGMP dependent Protein Kinase Ⅱ, PKG Ⅱ) normal tissue in the human digestive tract and its corresponding gastrointestinal expression in primary malignant tumors of PKG Ⅱ in tumor development of the role of clinical significance. Research methods: (1) In this study, immunohistochemical SP method PKG Ⅱ expression in 220 cases of normal human digestive tract tissue and their corresponding malignant tumors. (2) In this study, reverse transcription polymerase chain reaction (RT-PCR) assay PKG Ⅱ expression in 50 cases of normal human gastric tissue and 50 cases of primary gastric cancer tissues; application of reverse transcription-polymerase chain reaction (RT-PCR ) assay PKG II expression in 30 cases of normal human esophageal tissue and 30 cases of primary esophageal cancer tissues. (3) the relationship between clinical data and experimental results were analyzed. Results: (1) immunohistochemistry results: 1 110 patients with gastrointestinal cancer tissue PKG Ⅱ to express negative rate was 72.73% (80/110), corresponds to 110 cases of gastrointestinal normal tissue expression of PKG Ⅱ negative rate was 10.00% (11/110) . (2) of which 50 cases of gastric cancer organization PKG Ⅱ expression negative rate was 80.00% (40/50), 50 cases of normal gastric tissue PKG Ⅱ negative expression rate of 6.00% (3/50); PKG Ⅱ, 30 cases of esophageal cancer tissue expressing a negative rate of 56.67% ( 17/30), 30 cases of normal esophageal tissue PKG II negative expression rate of 3.33% (1/30); PKG Ⅱ, 18 cases of colon cancer tissue expressing negative rate of 72.22% (13/18), 18 cases of normal colon tissue PKG Ⅱ negative expression rate 22.22% (4/18); PKG Ⅱ, 12 cases of rectal tissue expression negative rate was 83.33% (10/12), 12 cases of normal rectal tissue PKG Ⅱ negative expression rate was 25.00% (3/12). (3) in which 10 patients with well-differentiated gastric cancer PKG II expression negative rate of 40.00% (4/10), 16 cases of differentiated gastric cancer PKG Ⅱ to express negative rate was 81.25% (13/16), 24 cases of poorly differentiated gastric cancer PKG Ⅱ negative expression rate of 95.83% (23/24), between the various degrees of differentiation of gastric cancer, PKG II-negative expression rates were significantly different (P <0.05); 11 patients with well-differentiated esophageal cancer PKG II expression negative rate of 27.27% (3/11), 9 patients differentiated esophageal cancer PKG Ⅱ expression of negative 66.67% (6/9), 10 patients with poorly differentiated esophageal cancer PKG Ⅱ negative expression rate of 80.00% (8/10). The degree of differentiation between esophageal cancer, PKG II negative expression rates were significantly different (P <0.05). ④ 21 patients with early gastric cancer PKG II expression negative rate was 71.43% (15/21), 29 patients with advanced gastric cancer PKG II expression negative rate was 86.21% (25/29), between the different clinical stages of gastric cancer, the PKG II negative expression rate not significant difference (P> 0.05). Negative rate was 64.29% (9/14), 16 cases of early esophageal cancer PKG Ⅱ expression negative rate was 50.00% (8/16), 14 patients with advanced esophageal cancer PKG Ⅱ expression between the different clinical stages of gastric cancer, PKG II negative expression rate was not significant difference (P> 0.05). (2) reverse transcription-polymerase chain reaction (RT-PCR) test results: normal gastric tissue group PKG II / GAPDH ((?) ± S) 1.532 ± 0.285, the primary gastric group the PKG II / GAPDH ((?) ± S) 0.456 ± 0.239, (P <0.01), were significantly different; the normal esophagus organizations group PKG II / GAPDH ((?) ± S) 1.841 ± 0.314, primary esophageal cancer group to 0.392 ± 0.212 ((?) ± S), (P <0.01) were significantly different. Conclusion: PKG Ⅱ expression in primary malignant tumors of the digestive tract were lower than normal gastrointestinal tissue. Primary gastric cancer, PKG II of expression and the degree of tumor differentiation, the lower the degree of differentiation, the lower PKG II expression: expression and age, gender, clinical stage, lymph node metastasis. Primary esophageal cancer, PKG II of expression and the degree of tumor differentiation, the lower the degree of differentiation, the lower the expression of PKG II: primary esophageal cancer, PKG II correlated with lymph node metastasis, lymph node metastasis PKG II expression, but its expression with age, gender, clinical stage. PKG Ⅱ reduction, deletions may be an important factor in the development of primary gastrointestinal malignancy, is a sign of deterioration of gastric cancer and esophageal cancer.
