Abstract


Purpose: CDX-2 LI-cadherin expression in gastric cancer and precancerous lesions, and the expression and distribution in cancerous gastric epithelial still unclear, through the study of gastric cancer tissues, adjacent tissues and normal mucosal tissue, gastrointestinal stromal tumors, chronic gastritis and intestinal metaplasia of the gastric mucosa tissue CDX-2 expression and gastric cancer patients of different gender and LI-cadherin expression levels and distribution, as well as the expression of the two positive rate differences with different types of gastric cancer, cell differentiation, depth of invasion of the lymph node metastasis of gastric cancer, gastric's Laurien staging and the clinical stage, study the relationship between the two in gastric cancer and precancerous lesions explore gastric precancerous lesions and cancer of the molecular mechanisms. Methods: application of the ready-to-use mouse monoclonal antibody (CDX2-88) and goat polyclonal antibody (C-17, sc-6987) using SP immunohistochemical detection of 246 cases of gastric tissue (including 30 cases of normal gastric mucosa, chronic superficial gastritis 30 cases, 42 cases of chronic atrophic gastritis, intestinal metaplasia of the gastric mucosa in 58 cases, 46 cases of gastric adenocarcinoma, 30 cases of cancer tissue, gastric stromal tumors 10 cases) CDX-2 and LI-cadherin protein expression situation. : CDX-2 gene protein expression in normal gastric mucosa and chronic gastritis; intestinal metaplasia positive expression rate was 91.4% (53/58); positive expression in gastric carcinoma was 80.4% (37/46 ), Laurien typing, intestinal type gastric cancer CDX-2 protein positive expression rate of diffuse type gastric cancer, and a significant difference (P lt; 0.050); CDX-2 cell differentiation are highly correlated (P lt ; 0.005), but with sex, age, depth of invasion, gastric cancer staging and lymph node metastasis. CDX-2 in cancer tissue and no expression in gastric stromal tumors. LI-cadherin gene protein expression in normal gastric mucosa and chronic gastritis; intestinal metaplasia positive expression rate was 83% (48/58); positive expression in gastric adenocarcinoma was 65% (30/46) press the Laurien points, intestinal type gastric LI-cadherin protein positive expression rate (78%) higher than the diffuse-type gastric cancer (35%), the difference was statistically significant (P lt; 0.050); LI-cadherin and lymph node metastasis and The clinical stage was positively correlated (P lt; 0.010), and in the adjacent tissues and no expression in gastric stromal tumors. CDX-2 over-expression of LI-cadherin are closely related, and CDX-2 LI-cadherin in intestinal metaplasia of the gastric mucosa showed strong accompanied relationship. Conclusion: The abnormal expression of CDX-2 may occur with early gastric, and may be a tumor suppressor gene, CDX-2 high expression of gastric low invasiveness, cell differentiation, and thus to detect the expression of strength to help determine the degree of malignancy of gastric cancer and the prognosis of patients with gastric cancer. LI-cadherin abnormal expression related to the occurrence of intestinal metaplasia and gastric adenocarcinoma, LI-cadherin high expression of the high rate of lymph node metastasis, and to detect the expression of strength can help determine the degree of malignancy of gastric cancer and the prognosis of patients with gastric cancer. CDX-2 and the LI-cadherine are intestinal-specific markers, and both expressed under normal physiological conditions in the small intestine and colon mucosa. This study shows that gastric intestinal metaplasia and gastric adenocarcinoma CDX-2 to adjust the LI-cadherine expression level, which is induced by the former, especially in the early stages of cancer tissue, and they may play an important cancer-promoting role . Cancer local invasion or distant metastasis is often the main reason for the clinical treatment failure, the molecular mechanism and biological behavior of gastric cancer incidence for the find new antimetastatic of measures is very critical.
