Abstract: Objective: To investigate the phytoestrogen daidzein (daidzein) in SD rats penis, testis and epididymis growth, development, and sex hormone receptors in rat corpus cavernosum and thus resulting effects on erectile function. Methods: 10 weeks (early adulthood), and 4 weeks of age (puberty) SD of 30 male rats were divided into five groups: normal control group, positive control group, low, medium and high doses of soy isoflavone groups 6, were given saline, 0.1mg / d DES (diethylstilbestrol, DES), 2mg, 20mg, 100mg/kg/d daidzein intragastric administration 90d, observed changes in body weight, in each group of rats testis, epididymis and penis index, HE staining testis, epididymis, and penile tissue structure changes; 5 of adolescent rats were measured in blood testosterone, LH and FSH levels, frozen sections prepared the penis, phalloidin-TRITC staining, laser confocal microscopy penile smooth muscle and collagen tissue distribution, RT-PCR method observed in each group sex hormone receptor mRNA in rat corpus cavernosum volume changes, immunohistochemistry and WestemBlotting observed penis sex hormone receptor expression changes. Results: The rats in early adulthood, low, medium and high dose groups of SD rats daidzein weight and penis, testis and epididymis index and normal control group had no significant difference (P all gt; O.05). In adolescent rats, the low, medium and high dose groups of the penis and epididymis index and normal control group had no significant difference (P gt; 0.05); testis index high-dose group (3.21 ± 0.07) was significantly lower than the normal control group ( 3.71 ± 0.32) (P lt; 0.01), however, the low-dose group and the normal control group, the difference was not statistically significant (P gt; 0.05); in the low dose group body weight and normal control group, no significant difference (P gt; 0.05), while the weight of the high dose group was significantly lower than the control group (P lt; 0.05); medium and high doses of testosterone in the blood of rats decreased penile smooth muscle tissue and collagen content ratio decreased, Compared with normal control group the difference was statistically significant (P lt; 0.05). Adolescence low, medium and high dose groups androgen receptor (androgenreceptor, AR), estrogen receptor α (estrogen receptorα, ERα) expression did not change significantly; low-dose group were estrogen receptor β (estrogen receptorβ, ERβ) expression did not change significantly, but the medium and high dose groups ERβ expression decreased. HE staining showed that adolescence and early adulthood, each dose soy isoflavone intake in rats on the penis, epididymis organizational structure had no significant effect, while the intake of high doses of soy isoflavone can stunted pubertal rat testis, there are different degrees spermatogenesis. Conclusion: early adulthood intake of daidzein on reproductive organs in rats had no significant effect. However, high doses of puberty (100mg/kg) of SD rat testis daidzein on the growth and development have a certain influence; low-dose daidzein (2mg/kg body weight) and development of adolescent rat penile smooth muscle and collagen content and erectile function no significant effect, but the intake of higher doses (≥ 20mg/kg) daidzein can cause penile smooth muscle tissue and collagen content ratio decreased, blood testosterone concentration decreased, penis ERβ expression is reduced, thus affecting erectile function of the penis .
