Abstract


Objective: To observe cutaneous squamous cell carcinoma (SCC) and normal skin tissue and the relationship between the expression of anti- apoptotic protein cFLIP cells , and explore its relationship with SCC clinical and pathological features and significance . Methods: Immunohistochemical the chemical affinity prime - biotin - peroxidase ( Avidin -Biotin - Peroxidase Complex , SABC ) detected 52 cases of cutaneous squamous cell carcinoma of the skin lesions and 13 normal controls cFLIP expression in skin tissue ; reverse transcriptase PCR (RT-PCR) and Western blot (western-blot) method for the detection of human skin squamous cell carcinoma cell lines A431 and immortalized the keratinocyte cells Hacat cFLIP expression . Results : The positive of cFLIP expression of cutaneous squamous cell carcinoma was 67.31% , while the positive rate was 15.38% in normal skin tissue , both express significant difference ( P lt; 0.01 ) ; of cFLIP expression and pathological grade , lymph node metastasis associated with gender , age , course of the disease , independent of the diseased parts ; of cFLIP and cell proliferation nuclear antigen (PCNA) expression in squamous cell carcinoma of the skin with a correlation ( r = 0.781 , P lt ; 0.01) ; human skin squamous cell carcinoma cell line A431 cFLIP mRNA and protein levels were higher than that immortalized keratinocytes forming cell lines Hacat . Conclusion : cFLIP expression in squamous cell carcinoma was significantly higher than the normal skin of cFLIP tumor pathological grade , lymph node metastasis associated with correlation and PCNA ; possible in the development of cutaneous squamous cell carcinoma play an important role . .
