Abstract


Objective: 1 . Prove that we build the pICOSIg of recombinant ICOS membrane outside the area and human IgG Fc fusion protein expression plasmid ; transient expression of the pICOSIg in COS7 cells and antigen-specific identification of product to lay the foundation for further study . 2 to explore pICOSIg stable expression in Chinese hamster ovary (CHO) cells and its expression product purified vitro preliminary detection of recombinant fusion protein 's biological function . 3 . Establish the rat small intestine transplant acute rejection in animal models to study the acute rejection period ICOS molecule expression in intestinal transplantation . The method : 1.Kpn I, and Bam HI digestion , the gene sequencing pICOSIg plasmid .
