Abstract


Medicinal fungi have a long history of use nutritional, medicinal and pharmacological properties. Their extracts, such as polysaccharide, protein etc, are widely sold as nutritional supplements and touted as beneficial for health, due to their medicinal effects, possibly including hepato-protection, antitumor, immuno-modulation. The work in this dissertation was based on submerged fermentation of Agaricus blazei Murill、 Ganoderma lucidum and Grifola frondosa to enrich the trace element in order to select a suitable carrier and a correct trace element. Agaricus blazei Murill and Zn were finally selectedThe Zn-accumulating、 Fe-accumulating 、 Se-accumulating characters of Ganoderma lucidum、 Grifola frondosa and Agaricus blazei Murrill were studied in this dissertation. The results showed that the capacity of Zn-accumulating、 Fe-accumulating、 Se-accumulating of Ganoderma lucidum 、 Grifola frondosa and Agaricus blazei Murrill were very strong. The best concentration of Zn、 Fe、 Se to Ganoderma lucidum was respectively 60ug/ml、 120ug/ml、 50ug/ml, The best concentration of Zn、 Fe、 Se to Grifola frondosa was respectively 30ug/ml、 100ug/ml、 30ug/ml, The best concentration of Zn、 Fe、 Se to Agaricus blazei Murrill was respectively 100ug/ml、 200ug/ml、 30ug/ml. The biotransformation extent of inorganic Zn into organic Zn of Ganoderma lucidum 、 Grifola frondosa 、 Agaricus blazei Murrill was respectively 77%、 60%、 55.5%, and that of Fe was 82%、 91% 、 82%.In this dissertation, It is compared the ability of bioenrichment of Zinc iron of Ganoderma lucidum 、 Grifola frondosa and Agarics blazei Murill, The results showed that the capacity of Zn-accumulating of Ganoderma lucidum was the strongest, when the Znic added to the liquid medium was up to 240mg/L, the hyphae have the most Znic absorbance, which was up to 30.39%;the capacity of Zn-accumulating of Grifola frondosa was the most weak, the most Znic absorbance of the hyphae is only up to 1.66%;the capacity of Zn-accumulating of Agarics blazei Murill was very strong, when the Znic added to the liquid medium was up to 200mg/L, the hyphae have the most Znic absorbance, which was up to 11.13%, so Ganoderma lucidum and Agarics blazei Murill can become organic carrier of Zn-enrichment. The antitumor activities on liver cancer in mice were assayed by tumor weight, all the fermenting liquor could show the antitumor activity and Agaricus blazei Murill was the most effective, its inhibition rate was 39.3%.The results showed that under the liquid culture conditions[pH5.6, fermention temperature at 25 ℃, the stirred speed 150r/min, inoculation size 15%, 250ml shake flash containing 50ml medium and in the medium containing Zinc], the Zn-recovery and the biomass of Agaricus blazei Murill were the highest. It is compared growth curve of submergedculture and Zn-accumulation culture. And orthogonal test of Zn> Se and Fe accumulation was completed: Zn 200 u g/mU SelO u g/mL, Fe200 u g/mL.Studies on polysaccharide isolation and purification process, intracellular polysaccharide was purified, AbMzn-l-A was the main fraction. Physics and chemical character showed that AbMzn-1-A is a a -D-Glucan structures polysaccharide, molecular weight 936771Da, have not linked protein or nucleic acid.
